Kate Winchester is John's older sister. She went missing in 1979 while trying to stop an evil witch. His fruitless efforts lead Sam and Dean down the same path when Sam finds an article matching his father's vague descriptions in his journal.

The brothers couldn't quite figure her out, and something about her left an impression they couldn't shake.

To cry uncle means to admit defeat and/or plead for mercy. It is not in Dean's nature, even in the face of insurmountable odds, to concede. When he stumbles across a situation with his niece, Mary, can his brother (and memories of Caleb's example with both himself and his boys) convince him that when it comes to raising Winchesters, sometimes you really do just need to cry uncle.
